BION-404

BION-404 is a suppressive recombinant protein for autoimmune disease modulation.

Scientific rational:

Endogenous BION-404 natural protein, is expressed and secreted by various Treg populations and suppress activated T,B and DC cells resulting in inhibition of auto-reactivity. Endogenous BION-404 axis is significant in maintaining peripharel tolerance. The immunosppressive role of the soluble form of human endodenous BION-404 was validated by Biond. BION-404 agents are recombinant monomeric fusion proteins that contains only a unique domain from the endogenous BION-404 protein

MoA:

Endogenous BION-404 binds the inhibitory FcyRIIB receptor expressed on immune cells and suppressess their activity, BION-404 lead agent acts as a monomeric, highly active immune cell suppressor with similar MOA to the endogenous FGL2.

BION-404 fusion proteins were generated and demonstrated; Binding to targer cells and receptors; Prevention of effector T cell proliferation and pro-inflammatory cytokines secretionl Inhibition of DC maturation and activation; Inhibition of the antigen-specific response of autoreactive T cells from mice.
